Abstract

: An important aspect in DNA base calling is to have readily available correlations between different variables involved to be able to make a more confident base call (identification of the DNA base) out of the raw data signal. Our previous work has shown a strong correlation between the spacing between consecutive bases and their type. In this way, we were able to create an algorithm for base calling that uses this information. In this paper, an additional such correlation is presented. It is shown that the amplitudes of the peaks from a DNA sequencing experiment are not completely random values and that there is a strong correlation between the type of consecutive bases and their relative peak amplitudes from the sequencing experiment.
